Un equipo de científicos de la Universidad de Oxford, en el Reino Unido, ha conseguido que ratones ciegos vuelvan a ver tras implantarles en sus ojos células que rehicieron la capa sensible a la luz de sus retinas, informa la Universidad de Oxford en un comunicado.Los autores del avance señalan que este método resultará clave en el tratamiento de pacientes con retinosis pigmentaria, un trastorno caracterizado por la degeneración progresiva de la estructura del ojo sensible a la luz, la retina, que poco a poco va perdiendo las principales células que la forman: los conos y los bastones. Este deterioro ocasiona una ceguera progresiva.El estudio fue dirigido por el profesor de Departamento Nuffield de neurociencia clínica de Oxford, Robert MacLaren y por Mandeep Singh, que es cirujano del Hospital Universitario Nacional de Singapur. Sus logros han aparecido detallados en la revista PNAS.

Recuperación de la sensibilidad a la luz Los investigadores trabajaron con ratones que estaban ciegos como consecuencia de la pérdida total de las células fotorreceptoras, sensibles a la luz, de sus retinas. Dos semanas después de que les fueran trasplantadas las células a estos animales, en sus ojos ya se había reconstruido por completo la capa detectora de luz de la retina, de tal manera que los ratones volvieron a ver.Las células empleadas fueron células ‘precursoras’ de ratón que se encontraban en la fase inicial hacia su desarrollo como células de retina.

En una prueba de constricción pupilar –un fenómeno ocular que se produce como respuesta a la iluminación-, se constató que de los 12 ratones que habían recibido el trasplante celular, 10 presentaban ante los estímulos lumínicos una construcción pupilar mejorada. Este efecto demostró que las retinas de los animales volvían a ser sensibles a la luz, y que las señales lumínicas estaban siendo transmitidas de nuevo a través del nervio óptico hasta sus cerebros.

A este respecto, Singh explica que: “Hemos descubierto que si trasplantamos a la vez un número suficiente de células, estas no solo se vuelven sensibles a la luz sino que también regeneran las conexiones necesarias para una visión significativa”.MacLaren añade: “Las células madre ya habían sido probadas en pacientes como medio de sustitución del epitelio pigmentario retinal o EPR(capa de células pigmentadas del exterior de la retina que nutre sus células visuales), pero esta nueva investigación demuestra que la capa retinal sensible a la luz también puede ser sustituida de forma similar. Las células sensibles a la luz presentan una estructura altamente compleja y hemos observado que pueden reactivar la función como capa y restaurar las conexiones, tras su trasplante en una retina completamente ciega”.

En lo que se refiere a la aplicación de este método para el tratamiento de la ceguera en humanos, MacLaren afirma que podrían usarse células madre pluripotentes inducidas o iPS, que son células madre que han sido generadas a partir de las propias células del paciente (de la piel o de la sangre), y que estas podrían ser dirigidas para formar precursores de células retinales. Esto ya ha sido logrado por otros investigadores, afirma MacLaren. El siguiente paso sería encontrar en los pacientes una fuente fiable de células, susceptibles de proporcionar células madre adecuadas para este tipo de trasplantes.

MacLaren concluye que, por ahora, lo que se ha conseguido es demostrar que “las células trasplantadas sobreviven, se convierten en sensibles a la luz y conectan y reconstruyen las conexiones al resto de la retina, para restaurar la visión. La capacidad de reconstruir por completo la capa retinal sensible a la luz es el objetivo final de los tratamientos con células madre para la ceguera, en los que actualmente trabajamos”.
